GST fusion protein

GST fusion protein
One way of purifying proteins expressed by a cloned gene is to insert the gene of interest into a vector in-frame with a gene encoding glutathione S-transferase; this is then expressed in a cell line to produce a (chimeric) fusion protein. This can be purified on a glutathione affinity column by virtue of the affinity of GST for glutathione, eluted with free glutathione and the GST can subsequently be proteolytically cleaved off and removed by an iteration of the affinity purification - though this is not always necessary.
